Platelet Depleted Plasma, Spectra APTT and Low Molecular Weight Heparin Evaluation of Propyl Gallate APTT to Monitor Low Molecular Weight Heparin Using Platelet Depleted Plasma By R Speck, D Bonderman, C Speck and J Griep Nov 15, 1996 Analytical Control Systems, Inc. has developed a new, patented highly sensitive Activated Partial Thromboplastin Time (APTT) assay. A new plasma preparation, platelet depleted plasma (PDP), is used for this test. Filtering platelet poor plasma (PPP) with a 0.22 micron double Millipore filter depletes the plasma of virtually all platelets and platelet fragments, increasing the sensitivity of the assay. A small, known concentration of phospholipid (PL) is added to the propyl gallate (PG) APTT reagent. The concentration of PL to optimize the sensitivity of this test has been determined. The effect of addition of various amounts of PL to the propyl gallate APTT for monitoring low molecular weight (LMW) heparin is shown below.

The two major outcomes of this research is decreased variability in the new plasma preparation technique and improved sensitivity of the propyl gallate APTT to both unfractionated and LMW heparin.
