Platelet Aggregation Assay (PAA) and Slide Platelet Aggregation Test (SPAT) activate platelets in a fashion unique among current aggregation agonist Mechanisms of Platelet Activation by Propyl Gallate: SPAT & PAA Propyl gallates mechanism of activation was investigated and in order to determine propyl gallates sensitivity, known platelet inhibitors were run on the platelet aggregometer. The mechanism of platelet aggregation assay (PAA) induced platelet stimulation was investigated. Luciferin/luciferase studies showed that PAA stimulated release of platelet granules. Platelet aggregation with 30 uM PAA was: blocked or severely inhibited by 0.025% Na Azide which blocks cytochrome B; partial inhibition was observed with 0.2mM RGD peptide which blocks platelet fibrinogen binding (no effect was observed with nonsense peptide); blocked with 1 uM sphingosine, an inhibitor of protein kinase C; blocked with 1 uM PGE1, an indirect stimulator of adenylate cyclase; and blocked with ASA, an inhibitor of cyclooxygenase.
